ABSTRACT:
Grapiprant (CJ-023423) is a new piprant-class non-steroidal anti-inflammatory medication that reduces pain and inflammation without inhibiting cyclooxygenase (COX) enzymes by specifically antagonising prostaglandin E2 (PGE2) EP4 receptors. Compared to traditional NSAIDs, this selective mechanism offers a better safety profile while maintaining protective prostaglandins. Grapiprant has shown encouraging therapeutic promise in both veterinary and human research, and it is mostly recommended for the treatment of osteoarthritis in dogs. The literature claims that a number of analytical techniques for Grapiprant estimation have been documented. As a result, the current evaluation gathers and condenses the published analytical techniques into a single, thorough source. High-Performance Liquid Chromatography (HPLC), Liquid Chromatography–Tandem Mass Spectrometry (LC–MS/MS), spectrofluorimetric techniques, and Reverse Phase-High-Performance Liquid Chromatography (RP-HPLC), including stability-indicating assay methods for method development and validation, have all been used to analyse grapiprant. This review primarily focuses on chromatographic techniques, especially RP-HPLC, which is still the most popular analytical method because of its accuracy, precision, resilience, and suitability for regular quality control examination. We go into great detail about important aspects of method development, such as detection wavelength, flow rate, mobile phase composition, and column selection. Additionally, in compliance with the standards set by the International Council for Harmonisation (ICH) and other regulatory bodies, crucial validation parameters like specificity, linearity, accuracy, precision, robustness, limit of detection (LOD), limit of quantification (LOQ), and system suitability are assessed.
