ABSTRACT:
Rutin, a plant derived flavonoid with varied pharmacological and medicinal qualities, was successfully detected using a cost effective UV spectrophotometric method developed through the Quality by Design approach. To ensure sustainability, rutin was extracted from the leaves and roots of Eucalyptus globulus employing ultrasound assisted extraction with eco friendly solvents, highlighting the integration of green chemistry principles. The UV method was systematically optimized via QbD to achieve robustness, reproducibility, and environmental compatibility. Method validation was conducted in compliance with ICH Q2(R1) guidelines, covering linearity, accuracy, precision, robustness, and ruggedness, thereby validating its stability for routine analysis of herbal extracts. The developed method exhibited high degree of linearity across the concentration range of 10–50µg/mL with a correlation coefficient of 0.997. Sensitivity parameters were established, with the limit of detection and limit of quantification determined as 0.82µg/mL and 2.48µg/mL, respectively. Precision studies revealed low relative standard deviation values, while robustness and ruggedness assessments confirmed the reliability of the method under varied analytical conditions. The validated procedure was successfully applied for the detection and estimation of rutin in Eucalyptus globulus leaf and root extracts, demonstrating its practical utility in herbal formulations. Overall, this study establishes a novel, simple, precise, and sustainable Quality by design based UV spectrophotometric method for rutin detection. The integration of green extraction with validated analytical performance underscores its effectiveness for routine quality control and research applications in phytopharmaceuticals.
